incapacitating consequences like movement dysfunction, cognitive decline, and neurological
impairments. It is essential to manage stroke effectively to lessen its negative effects and
enhance patient outcomes. The potential neuroprotective benefits of acetylcholinesterase
inhibitors may assist maintain the integrity of brain tissue and lessen the damage that neurons
may sustain after a stroke, making them a promising option in this situation. This
comprehensive thesis explores the neuroprotective benefits of an acetylcholinesterase inhibitor,
Rivastigmine, in relation to stroke outcomes in a variety of ways. Using an advanced
integrative methodology, this study includes behavioral evaluations, in silico analysis,
histopathological results, and molecular studies to give a comprehensive picture of the possible
mechanisms behind therapeutic effects of Rivastigmine. Within the field of computer
modelling, the docking interactions between SOD2 and TLR4, the target proteins of
Rivastigmine, provide detailed structural information that directs further experimental
validations. Following Rivastigmine treatment, the behavioral tests conducted exhibited
improvements in cognitive, motor, and social domains. These results were further corroborated
by histopathological analysis, which shows neuroprotective effects in Rivastigmine treated
group as opposed to surgery (MCAO) group. Post Rivastigmine treatment, real-time PCR data
showed a rise in SOD2 and a decrease in TLR4 levels in surgery (MCAO) rats, exhibiting
antioxidant and anti-inflammatory effects of Rivastigmine. These findings provide interesting
directions for future neuroprotective approaches and shed light on the possible therapeutic
implications of Rivastigmine in reducing neurodegeneration that occurs in stroke.

                        , Heat treatment plays a major role in improving the material properties. With the
addition of cryogenic treatment these properties can be further improved. In this paper heat
treatment is carried out on low carbon low alloy structural steel in two different types of
furnaces conventional box type air furnace and vacuum furnace along with shallow cryogenic
treatment with the purpose to investigate the outcome of shallow cryogenic treatment on the
material properties and to find out which type of furnace is more suitable with shallow
cryogenic treatment. For this purpose, structural steel 30CrMnSiA was selected, and heat
treatment was carried out in conventional box type air furnace (900°C × 40mins) and vacuum
furnaces (900°C × 70mins), shallow cryogenic treatment (-75°C × 125mins) was carried out
after the quenching and before tempering. For the physical properties of the material,
hardness (HRC) was investigated using Brinell hardness tester HBD-62-5AP. Tensile
samples were prepared as per ASTM E8/E8M and investigated using 100kN WAW-100B
Electro-Hydraulic Servo Computer Control Universal Testing Machine. Impact toughness
was measured by using electronic pendulum impact testing machine CBD-300 and specimens
were prepared using ASTM standard E23-12c. Microstructural analysis for grain structure
was carried out using Olympus DSX1000 optical microscope.

                        , Cancer and its reoccurrence have become a major health problem affecting the
quality of life for millions of individuals every year. The currently available strategies,
radiotherapy, and chemotherapy have non-specific targets, lower solubility, and severe
side effects. Hydrogels are crosslinked polymeric networks with higher swelling
properties, degradation, biocompatibility, and flexibility which can be manipulated based
on the desired application. Incorporating AV can elevate the antioxidant, and anticancer
properties of the hydrogel based drug delivery system (DDS) also warranting enhanced
swelling, drug release, and environmental degradability of the system. Acrylic acid (AA)
induces pH-responsive properties in the hydrogels. The PVA/SA and PVA/SA/AV
hydrogels were synthesized with a pH responsive behavior and investigated at different
pH conditions. The unloaded and loaded PVA/SA and PVA/SA/AV hydrogels with
imatinib (IM) were characterized for their structural morphology, physiochemical
characteristics, and antioxidant and anticancer activity. The IM loaded PVA/SA/AV
hydrogels showed increased pore size in SEM micrographs, enhanced swelling abilities
up to 400%, 100% degradation, 56% encapsulation efficiency, and drug release profiles
of up to 94% in 24 h, compared to PVA/SA hydrogels loaded with IM. Dpph radical
scavenging activity was also observed to be enhanced in PVA/SA/AV hydrogels. Finally,
the cell viability analysis in resistant MCF-7 breast cancer cell lines exhibited 41% cell
viability of the PVA/SA/AV hydrogels loaded with IM implying a promising potential of
AV to be incorporated in a hydrogel based drug delivery system for targeting drug
resistance in cancer.

the major barriers in diabetic wound healing, specifically targeting chronic inflammation,
oxidative stress, bacterial infection, and impaired angiogenesis. This therapeutic system integrates
cerium oxide nanoparticles (CeO₂), metal-organic frameworks (MOFs), and a chitosan-gelatin
hydrogel platform to create a multifunctional wound dressing. CeO₂ nanoparticles provide strong
antioxidant and antibacterial activity, MOFs enable controlled drug release and add antimicrobial
properties, while the chitosan-gelatin hydrogel ensures biocompatibility, moisture retention, and
accelerated tissue repair. Comprehensive characterization, including Raman spectroscopy, FTIR,
UV-Vis, EDX analysis, and optical microscopy, confirmed successful synthesis, structural
integrity, and the presence of desired functional groups. Biological assays demonstrated superior
antibacterial and antibiofilm performance of the CeZn-Gel composite against E. coli and S. aureus,
surpassing individual system components and controls. The integrated approach not only enhances
wound healing outcomes but also reduces infection and inflammation, with significant potential
for improving patient quality of life in chronic wound care. Future work will focus on optimizing
formulation and advancing toward clinical testing for broader therapeutic impact.

                        , Karyotyping continues to be the bedrock of cytogenetic diagnosis, providing key information
on chromosomal abnormalities causative of a broad range of genetic disorders, developmental
abnormalities, and cancers. But standard karyotyping is time-consuming, requires extensive
specialist interpretation, and is vulnerable to human mistake and inefficiency, especially in
high-volume clinical settings. Despite advances in medical image analysis, the automation of
karyotyping faces persistent challenges, including the lack of large-scale annotated datasets,
difficulties in segmenting overlapping chromosomes, and variability in chromosome
morphology and staining. These challenges define a significant research gap in developing
scalable, accurate, and clinically deployable deep learning models for automated chromosome
analysis.
In order to overcome this gap, we first present a large-scale, clinically annotated cytogenetic
database, built from 1,311 patients and consisting of 10,057 karyograms with 514,949
manually annotated chromosome singlets. Furthermore, 3,935 metaphase images are annotated
at the instance level in COCO format. This data set reflects true-world diversity, ranging from
normal and abnormal karyotypes to different Giemsa (G-banding) staining intensities,
structural abnormalities, and overlapping difficult cases, and thus presents a solid basis for the
creation and testing of deep learning models within automated cytogenetics.
Based on this work, we create two primary methodologies aimed at the fundamental tasks of
karyotyping. For segmentation of chromosomes, we introduce a variant Mask R-CNN model
involving an Attention-based Feature Pyramid Network (AttFPN), spatial attention, and a
LastLevelMaxPool component to improve multi-scale feature representation and contextual
perception. It enhances performance in difficult situations, including overlapping
chromosomes and weak banding patterns, and gains considerable improvements in mean
Average Precision (mAP) compared to standard baselines.
For chromosome classification, we present the Dual Attention Multiscale Pyramid Network
(DAMP), a specifically designed model that combines channel and spatial attention
mechanisms to concentrate on discriminative features, as well as a multiscale pyramid
architecture to cope with size, orientation, and quality variation in chromosomes. DAMP's
highest classification accuracy is 96.76% on both public and commercial datasets, performingxv
better than state-of-the-art models like ResNet-50, Vision Transformers, and Siamese
Networks.
Overall, this thesis provides interpretable and scalable deep learning models for automating
chromosome classification and segmentation. Through the closure of key gaps in dataset
quality, model resilience, and clinical utility, the work facilitates the insertion of clever
decision-support systems into cytogenetic pipelines, ultimately leading to improved diagnostic
reliability and efficiency in the face of chromosomal disorders.

                        , Total Knee Replacement (TKR) surgeries are becoming increasingly common globally as
an effective measure to counter knee arthritis. Total knee replacement implants are very
advantageous in a sense that they offer 99% success rate to patients. This thesis presents
the design, simulation and additive manufacturing of a patient specific, Functionally
Graded Lattice Structure (FGLS) knee implant in Ti 6Al-4V alloy with specific reference
to the healthcare situation in Pakistan. This was to explore the local manufacturing facilities
of Pakistan as all knee implants are imported from abroad.
The strategy involved a high degree of workflow consisting of Computer-Aided Design
(CAD), finite element analysis (FEA), and topology optimization using nTopology to
create Gyroid-based lattice work. The structures were to resemble the trabecular bone
structure to ensure that stiffness discrepancies were minimized. This helped counter only
one drawback of solid knee implants, stress shielding.
The simulations of the physiological loading conditions (static and cyclic) demonstrated a
Von Mises peak of 620.45 Mpa and safety factor of 12.66 on the average and unlimited
predicted life of fatigue of over 10^7 cycles. The use of FGLS was effective in making the
weight of 490 g to 292, leading to a 40 percent weight reduction, with no structural integrity
lost. Selective Laser Melding (SLM) was used to fabricate the implant and the heat
treatment allowed stress relieving of the additively manufactured implant.
Compressive testing was also mechanically vindicated to be on an of average 95.02 kN
with little variation and Micro-CT scanning confirmed high dimensional fidelity and
showed internal lattice geometries without defects. According to this research, SLM
produced FGLS implants usage has proven to be an option to traditional prosthetics, which
is mechanically stable, biologically desirable, and cost-effective, and has a bright future of
being a locally manufactured orthopedics product.
